A microfluidic integration of planar patch-clamp chip with rapid solution exchange system and optical detection

Abstract

This work presents an integration of planar patch-clamp chip with microfluidic solution exchange system and optical measurement for ion-channel recording. Perforated patch for whole-cell recording was implemented via intracellular solution exchange. Dection of temporal fluorescent variance during extracellular solution exchange was also demonstrated. The increased fluorescent intensity indicates both the usefulness of extracellular solution exchange and the ability of continuously optical observation. Conceivably, the device can be further applied to simultaneous electrical and optical measurement for cellular physiological study.
