Electrooxidation of NADH with Co(phen)33+ by NADH dehydrogenase purified from Bacillus stearothermophilus

Abstract

The NADH dehydrogenase purified from Bacillus stearothermophilus catalyzes effectively the electrochemical oxidation of NADH mediated by Co(phen)3+3. Stopped-flow measurements indicate that the rate constant for the oxidation of the reduced enzyme by Co(phen)3+3 is 5 × 106 M-1 s-1 and that for the oxidation of NADH by the oxidized enzyme > 3 × 107 M-1 s-1. The voltammograms, showing catalytic prepeaks, were simulated using the above rate constants.
