The study of changes of the mitochondrial respiratory chain enzymes in the bbn induced bladder tumor in rat

Abstract

To investigate the mitochondrial respiratory chain enzymes change in the process of tumorigenesis of bladder tumor in rat. BBN was administered orally to the male Spraque-Dawley rats with the concentration of 0.05% for 8 weeks, then the urinary bladder was harvested periodically for evaluation of tumorigenesis and preparation of the mitochondrial suspensions for the measurement of respiratory chain enzyme activity; the examined enzymes included NADH cytochrome c reductase (NCCR) , succinate cytochrome c reductase (SCCR) and cytochrome c reductase (CCO). The induced bladder tumors are transitional cell carcinoma and mainly superficial and low grade in pathology, the enzyme activity changed significantly since the 10th week after oral administration in NCCR & SCCR and since 14th week in CCO during the period of observation, the SCCR activity in tumor tissue is significantly different from that in normal one. These revealed the mitochondrial enzyme activity is changed during the induction of tumor and is different in the tumor tissues as compared with those of normal tissues.
