Fabrication and cell affinity of poly(vinyl alcohol) nanofibers via electrospinning

The nanofibers of poly(vinyl alcohol) prepared in this paper were fabricated by electrospinning and characterized using scanning electron microscopy (SEM), Brookfield Rheometer and atomic force microscopy (AFM). In order to observe cell morphology and proliferation on electrospun poly(vinyl alcohol) nanofiber for tissue engineering applications, fibroblast cells were seeded onto the poly(vinyl alcohol) scaffold and grown in incubator. Inverted microscopy and SEM were used to observe cell morphology and proliferation. A concentration of 4% poly(vinyl alcohol) was too low to form uniform nanofiber at applied voltage from 15,000 V to 25,000 V. The viscosity of 12% poly(vinyl alcohol) was quite high. Therefore, for 12% poly(vinyl alcohol), it is necessary to create a strong electric field (higher voltage) to draw nanofiber. The best processing condition for fabricating uniform poly(vinyl alcohol) nanofiber in this study was 8% in concentration at applied voltage of 15,000 V. Based on this condition, the nanofibrous scaffold was prepared for fibroblasts to seed on it. The fibroblast cells grew significantly after 7 days' culture, but the attached fibroblasts were round-shaped on the PVA nanofiber scaffold.
