N2 - This study demonstrates an optimized electroporation (EP) system for gene transfection by the Taguchi method. The parameters of the EP system, having an influence on the transfection rate, were optimized, resulting in optimized values of 50 μm electrode gap, 80 μg/mL pEGFP-N1 concentration, 6 V pulse voltage, and 2-pulse per time with above 95% confidence. The results show that the gene transfection efficiency is highly enhanced due to the dramatically increased pEGFP-N1 concentration by the electrostatic force. With optimized factors, the pEGFP-N1 plasmid was successfully transfected into the basal cell carcinoma cell line (BCC) with 35.89 % average transfection rate.
