Positively charged GFP-chromophore-based fluorescent sensor for DNA

摘要

Fluorescence titration of the positively charged green fluorescent protein (GFP) chromophore analogue 1 with E. Coli plasmid DNA [pcDNA3.1(−)] confirms that 1 serves as a fluorescent probe for sensing E. Coli plasmid DNA. The binding constant K between the DNA and 1 is as large as 2.9 × 105. The sensing mechanism is that the intermolecular steric hindrance generated between 1 and E. Coli plasmid DNA by electrostatic attraction restricts the φ-torsion around P-bond and τ-torsion around I-bond of 1, enhancing fluorescence of 1.
