Since traditional Chinese medicines can effectively relieve symptoms of asthma the agents have been used as oral medications for anti-asthma in Asian countries within centuries In this study we showed a novel medication by developing of a complex traditional Chinese medicine (CTCM) in nebulized form so called as OTM66 OTM66 was applied through the intratracheal route and found to be effective against an early-phase air-flow obstruction in ovalbumin (OVA)-sensitized guinea pigs The application of OTM66 inhibited IL-5 production in OVA-sensitized guinea pigs We further examined the mechanism of OTM66 to determine its function in the early-phase response to OVA challenge Using we found that the inhibition of bronchoconstriction by OTM66 was attenuated by pretreatment with the β-adrenergic antagonist propranolol suggesting that OTM66 exerts a bronchodilator effect via the β-adrenergic receptor-dependent pathway Moreover OTM66 was able to inhibit histamine and IL-4 secretion in the OVA-induced airway hypersensitivity in guinea pigs at 5 min post-OVA challenge The in vitro study using P815 revealed that treatment with OTM66 stabilized mast cells In conclusion our results indicate that OTM66 is a promising bronchodilator and works well as a mast cell stabilizer The findings provide important evidence showing its application for the treatment of allergenic airway disorders
